Even though I love Bewitched, I've never considered Darrin and Larry best friends (although that's what they called each other). I mean, what kind of "best friend" would constantly fire (or threaten to fire) you or berate you over the most simple mistakes?
I do agree with Majors Nelson and Healy, as well as Andy and Barney.

Hal Walters and Stanley Beamish (Mr. Terrific)
I grew up thinking that they had to be the very best of friends. Now, forty years later, I have to admit that, personality wise, my best friend is very much a Hal and I'm a Stanley.
If Gilligan and the Skipper were Laurel and Hardy, (In many ways they were. Alan Hale did "Ollie" takes and gestures throughout the series.) then Hal and Stanley were Martin and Lewis. (Though Stanley was "brighter" than Jerry's character.)
Recently I e-mailed Dick Gautier (Hal) and asked about their relationship. It was a little disheartening to learn that he did not get along with Stephen Strimpell (Stanley) at all. But now that I've had the chance to view several episodes again, I must say that they both did a wonderful job of "acting" like friends.

Batman & Robin - Batman (http://www.sitcomsonline.com/boards/forumdisplay.php?f=363)
I'd go with Andy & Barney. Then the Skipper & Gilligan would be a close second.

TV's most underrated female buddy was Laura Petrie and Millie Helper of The Dick Van Dyke Show. Mary Tyler Moore and Ann Morgan Guilbert did great comedy together and too bad more storylines were not done with them front and center.
OH Nuts!
