OK - this should be easy. The sitcom had a PUPPET as an old lady that ran a business. Ring any bells? This PUPPET was well known and she did the comedy circuit with her "handler"...

Sounds like a series called "Madame's Place". I think it ran in the early 80's.

Yeah the guy's name is called Wayland Flowers and the name of the old lady puppet was called Madame. I think I also saw them on the Hollywood Squares (late 70's early 80's)

Wayland Flowers was gay and eventually died of AIDS in the 1980s... that Madame puppet was famous and on many shows like Hollywood Squares, the variety shows... think it was even on DeaN Martin roasts...

Their biggest claim to fame was as regulars on the early 80s series "Solid Gold".

They were also on a summer replacement series that aired Saturdays on ABC. I can't remember the name of it, but the cast included Flowers, mimic Fred Travalino, Gailard Sartain & Jack Riley (Mr. Carlin from "Bob Newhart")
